Human CXCL14 (BRAK, breast and kidney expressed chemokine) is a CXC chemokine family member consisting of 77 amino acid residues in its mature, secreted form. An orphan chemokine ligand since its discovery in 1999, CXCL14 was recently described as a ligand for MAS-related GPCR X2 (MRGPRX2) and ACKR5. MRGPRX2 is expressed on mast cells and, like CXCL14, upregulated in bronchial inflammation. Structural studies of CXCL14 revealed a very short N-terminal sequence and an unusually long loop between the second and third beta strands that promotes ubiquitin mediated degradation.
